Phosphodiesterase-3 (PDE-3) Inhibitor
Mechanism of Action (MOA)
Inhibits PDE-3 → ↑ cAMP → vasodilation and antiplatelet effect; improves claudication walking distance.
Black Box Warning
Contraindicated in heart failure of any severity — PDE-3 inhibitors decrease survival in patients with HF.
Clinical Notes
Symptomatic relief of intermittent claudication in PAD. Headache, palpitations, diarrhea. Do not use in any degree of heart failure.
